High mobility group 1 (HMG1) is a highly conserved non- sequence- specific DNA-binding
nuclear protein. Mammalian HMG1 is composed with two homologous DNA-binding domains
HMG boxes, A and B, a short basic region and an acidic C-terminal domain containing 30
consecutive Asp and Glu residues. Recombinant human HMG1 was expressed in E coli and
purified by the combination of Niaffinity and conventional/FPLC chromatography methods to
>95% homogeneity. It has been tested for its activity in different assays including in vitro
transcription, gel mobility shift assay and TNFα-induction assay.
