Human Positive Cofcator 4 (PC4) was purified from HeLa cells and capable of enhancing activated transcription by diverse gene- or/and tissue-specific DNA-binding nuclear protein. Phosphorylation of PC4 at its N-terminal domain negatively regulates its ability to bind to activators and DNA. Recent studies have demonstrated that PC4 is one of the common cancer markers. Its expression correlates well with tumorigenesis of most common tumors. Recombinant human PC4 was expressed in E coli and purified by the combination of conventional and FPLC chromatography methods to >95% homogeneity. It has been tested for its activity in different assays including in vitro transcription, gel mobility shift assay and DNA-binding assay.
